Objective:To observe the influence of Persicae Semen on tubular erithelial myofibroblast transdifferentiation in rats with renal interstitial fibrosis induced by unilateral ureteral obstruction.Method: Rat model of unilateral ureteral obstruction was used.The rats were randomly divided into 4 groups:nomal group
mode group
monopril treatment group
Persicae Semen treatment group.Renal tissue was examined by light microscopy at the 2th week after operation.Immuno-histochemistry was applied to measure the expression of E-cadherin
α-SMA and FN in the renal interstitium.Result:Fibrosis was observed in the renal interstitium of rats with obstructive nephropathy
the expression of α-SMA and FN in the normal operation group was low in the renal tubular epithellal cell and renal interstitium cell
but the expression of E-cadherin was high.In comparison with normal group
the expression of α-SMA and FN of model group was markedly increased
but the expression of E-cadherin was decreased(P<0.05).In comparison with model group
Persicae Semen group could significantly decrease the expression of α-SMA and FN
but the expression of E-cadherin was increased(P<0.05);no significant difference was detected between Persicae Semen treatment group and monopril treatment group.Conclusion: Pesicae semen may suppress fibrosis development
at least in part via down regulating α-SMA and FN expression.
