目的:观察雷公藤提取物长期用药对大鼠心脏毒性的影响。方法:将SD大鼠随机分为空白对照组和雷公藤提取物低、中、高剂量组17

90 d测定各组血钾、心电图及心功能指标
并采用HE染色法观察大鼠心肌组织的病理变化。结果:低剂量组与给药前或空白对照组比较血钾、心电图及心功能各指标无明显变化。中剂量组给药后60~90 d出现血钾降低
心电图指标如Q-T间期、S-T间期、QRS间期延长、心率降低和ST段偏移
心功能指标如左室收缩压(LVSP)降低、左室舒张末压(LVEDP)升高、左室内压最大上升/下降速度(±dp/dtmax)降低(P<0.05)。高剂量组在给药后15~30 d上述指标已有明显变化(P<0.05)。HE染色显示
给药后90 d低剂量组大鼠心肌未明显见病理变化
中、高剂量组大鼠心肌有明显损伤。结论:雷公藤提取物连续ig给药90 d后
17 mg·kg-1剂量组对大鼠心脏无明显毒性作用

Objective:To observe the toxic effects of Tripterygium wilfordii extract on chronic heart injury in rats.Method: Sprague-Dawley rats were administrated and randomly divided into four experimental groups:control group
low dose group
middle dose group
and high dose group of T.wilfordii(17

the electrocardiogram
serum potassium and heart function were tested.The pathological changes were evaluated by Haematoxylin-eosin(HE).Result: There was no changes in electrocardiogram
serum potassium and heart function of the rat in low dose group compared with that of the control group.But in the middle dose group
on the 60th day
Q-T interval
S-T interval
QRS interval was prolonged
heart rate decreased significantly
S-T segment changed significantly compared with those of the control group.While potassium was decreased
left ventricular systolic pressure(LVSP)
maximum slop of left ventricular pressure(±dp/dtmax)was significantly reduced
and the left ventricular end diastolic pressure(LVEDP)was increased significantly(P<0.05).The high dose group had the similar changes with the middle group on 15th day after administrated(P<0.05).The middle dose and high dose of the T.wilfordii extract could induce significant pathological changes of rat heart compared with that of the low does group and the control group.Conclusion: The middle does and the high does of T.wilfordii extract could induce significantly cardiac injury
and pathological changes of cardiac muscular tissue.But low dose has no obvious toxicity for heart.
