Objective:To observe the effect of icariin(ICA) on the levels of matrix metalloprotease-2(MMP-2) in rat carotid artery injured by balloon and to evaluate the mechanisms of inhibiting intima hyperplasia and vascular remodeling.Method: Fourty-four rats were randomly divided into 4 groups: natural group
model group and ICA low dose group(60 mg·kg-1) and ICA high dose group(120 mg·kg-1).Rat carotid balloon injury restenosis model was established.Rats of ICA group were treated with(ig) drugs for 2 or 4 weeks.Then the mRNA expression of MMP-2 was assessed by RT-PCR
and immunohistochemistry analysis was applied to detect the expression of MMP-2.Result: Compared with model group
the ICA group significantly decreased the expression of MMP-2
at 14 and 28 day after injury(P<0.01).Conclusion: ICA could inhibit neointimal proliferation of rat carotid artery injured after balloon injury
the mechanism might be related to the effects of ICA in decreasing mRNA level of MMP-2
inhibiting migration of vascular smooth muscle cells.
