Objective:To study on separation and purification technology of andrographolide and dehydroandrographolide by macroporous resin.Method: Based on screening model of resin
characteristics of adsorbing and eluting parameter had been obtained by HPD100 resin
and the content of andrographolide and dehydroandrographolide were determined by HPLC.Result: Optimum parameter of purification were as follows: maximum sample volume was 1.0 g of resin per medicine material
5 BV 20% ethanol was used for eluting impurity
for eluting target components was 10 BV 35% ethanol(for andrographolide) and 5 BV 55% ethanol(for dehydroandrographolide).Purity of andrographolide and dehydroandrographolide were respectively 48.59% and 19.87% and total lactone was 68.46%.Conclusion: HPD100 resin was favorable to andrographolide and dehydroandrographolide under condition of determined process
and it had significance for large-scale production.
