此法可以测定27种氨基酸的含量
但不适合清开灵注射液中氨基酸的含量测定。结论:该方法不能对清开灵注射液中氨基酸进行准确定量。
Abstract
Objective: To develop a method for determination of amino acids by on-line derivatization.Method: The amino acids were analyzed by HPLC with orthophthalaldehyde(OPA) and 9-formic acid methyl ester of fluorene chlorine(FMOC) on line derivatization.The sample was analyzed on a Zorbax Eclipse-AAA column(4.6 mm×150 mm
5 μm) with gradient elution of 40 mmol·L-1 Na2HPO4(make pH be 7.8 with 8 mol·L-1 NaOH)(A) and acetonitrile-methanol-water(45∶ 45∶ 10)(B) at the flow rate of 2.0 mL·min-1
detected at 338 nm and 262 nm.Result: This method could be used to analyze 27 amino acids in the range of certain concentration
but it wasn’t fit for the determination of amino acids in Qingkailing injection.Conclusion: This method couldn’t determine amino acids in Qingkailing injection accurately.
