Objective:To optimize separation and purification technology of total saponins from Dioscorea bulbifera.Method: With the content of total saponins from D.bulbifera as index
which was determined by ultraviolet spectrophotometry
optimum macroporous resin model was selected by dynamic adsorption-desorption method
then separation and purification technology of total saponins from D.bulbifera was optimized.Result: Optimum separation and purification technology conditions of total saponins by D-101 macroporous resin were as follows:the concentration of extraction liquid from D.bulbifera 0.2 g · mL-1
sample volume of 6 BV
flow rate of 1.0 mL · min-1
washed impurity with 4 BV water
and then desorpted with 4 BV 30% ethano1
collected eluate.Under these technology conditions
yield of total saponins from D.bulbifera reached as high as 61.85%.Conclusion: This optimized technology was simple and feasible
