Objective:To study the synergic effects of Shouwu Fang and Madopar on striatal neurochemical substances in freely moving rats with Parkinson’s disease(PD) by brain microdialysis and its relevant detection technology.Method: SD rats were randomly divided into control group

Shouwu plus Madopar group.Parkinson’s disease animal model was induced by injection of 6-hydrodopamine(6-OHDA) into striatum of SD rats.Brain microdialysis was performed and concentrations of dopamine(DA) and its metabolites homovallinic acid(metabolic protact of DA)

5-dihydroxy benzyl acid(the index of hydroxyl radical) were detected by high performance liquid chromatography-electrochemical detector(HPLC-ED).Result: Compared with Madopar group
Shouwu Fang plus Madopar group treatment elevated extracellular DA level in striatum with peak at(39.15±28.70) μg · L-1 as well as reduced the fluctuation of DA(ΔDA).In addition
during whole observation period
hydroxyl free radical generation in Shouwu Fang plus Madopar group was significantly lower than Madopar group(with 9 time points P<0.05).Conclusion: Shouwu Fang could raise DA concentration and reduce its fluctuation in extracelluar fluids of straitum.It could also inhibit the generation of hydroxyl free radical and protect neurons from damage under oxidative stress.
