目的:比较白及块茎和须根的抑菌作用
并从中筛选出抑菌的有效部位。方法:采用滤纸片法和光电比浊法比较白及块茎和须根水提物及醇提物的抗菌活性
并进一步采用系统溶剂萃取法对其活性部位进行定位;最后
通过HPLC比较白及块茎和须根提取物的化学成分的差异。结果:白及块茎和须根的水提物基本无抑菌效果
而醇提物对多种细菌具有抑制活性
其中须根优于块茎;氯仿层、乙酸乙酯层和正丁醇层为其抑菌活性部位
其中须根乙酸乙酯层对枯草的抑制效果最好
在1 g.L-1的浓度下抑制率可达50.39%±3.53%。HPLC结果表明
须根和块茎化学成分基本一致
但单位含量前者显著高于后者。结论:白及须根和块茎抑菌活性成分主要位于脂溶性的醇提物中
两者化学成分相似
但须根中物质含量及抑菌效果相比较均优于块茎
具有一定开发应用价值。
Objective:To study the antimicrobial effect and screen antimicrobial effective parts of the tubers and fibrous roots of Bletilla striata.Method: Filter paper method and photoelectric turbidimetry method were carried out to evaluate the antibacterial effect of the extracts of the tubers and fibrous roots of B.striata.The system solvent extraction method was adopt to further location of the active sub-fractions.Finally
HPLC method was applied to compare the chemical difference between tubers and fibrous root extract of B.striata.Result: The aqueous extract of tubers and fibrous roots of B.striata had no inhibitory effect while the 95% ethanol extracts of both tubers and fibrous root showed antibacterial effect;sub-fraction analysis manifested that the chloroform layer
ethyl acetate and n-butanol layer were the active part of the tubers and fibrous
which the ethyl acetate layer of the fibrous roots with the strongest inhibitory activity.HPLC revealed that the chemical composition of 95% ethanol extraction of both fibrous roots and tubers of B.striata were similar
while the former had remarkable amounts of content in several components.Conclusion: The antibacterial active components were mainly deposit in the hydrophobic 95% ethanol extract for both fibrous roots and tubers of B.striata
and they were similar in chemical composition
while the former showed higher amount of chemical components and better antibacterial activity
which implied the fibrous roots of B.striata with potential value.
