Objective: To optimize successive ultrasound countercurrent extraction process of Ligustri Lucidi Fructus. Method: Taking total amount of oleanolic acid and ursolic acid as index
orthogonal design was adopted to investigate effects of ethanol concentration
ultrasonic frequency
solid-liquid ratio and temperature on extraction process of Ligustri Lucidi Fructus.ELSD was employed to determine contents of oleanolic acid and ursolic acid with mobile phase of acetonitrile-methanol-0.5% acetic acid(19:67:14) and flow rate of 0.33 mL·min-1. Result: Optimum extraction process was that extracted with 15 times the amount of 80% ethanol at 30 kHz; under these conditions
average total amount of oleanolic acid and ursolic acid was 5.66% with RSD of 1.64%. Conclusion: This optimized extraction technology was reasonable and scientific
which was suitable for large scale production of Ligustri Lucidi Fructus.
