Objective: To optimize β-cyclodextrin(β-CD) assisted extraction process of Polygoni Multiflori Radix Praeparata. Method: Analytic hierarchy process was adopted
taking comprehensive score of transfer rates of emodin
physcion and stilbene glycoside as index
orthogonal test was applied to optimize extraction technology with solid-liquid ratio
β-CD dosage
extracting time and times as factors.HPLC was employed to determine contents of emodin
physcion and stilbene glycoside with mobile phase of acetonitrile(A)-0.1% phosphoric acid solution(B) for gradient elution(0-5 min
15%-25%A;5-12 min
25%-30%A;12-15 min
30%-60%A;15-20 min
60%A;20-25 min
60%-75%A;25-31 min
75%-77%A) and detection wavelength at 320(stilbene glycoside) and 254(emodin and physcion) nm. Result: Optimal extraction process was as following:extracted 3 times with 10-fold water for 1 h each time
β-CD dosage of 10%;Transfer rates of emodin
physcion and stilbene glycoside were 8.39%
0.76% and 90.19%
respectively. Conclusion: Compared with traditional water extraction
β-CD assisted extraction process could significantly improve transfer rate of active ingredients.
