Objective: To extract and purity Astragali Radix glycoprotein(AmGP-2) having immunosuppressive activities
then investigate its physicochemical properties and composition. Method: AmGP-2 was handled by water extraction
salting separation
sevage method; Then purified by anion and gel chromatography.Physicochemical properties and composition of AmGP-2 were analyzed by phenol-sulfuric acid method
BCA method
Molish test
ninhydrin reaction
gas chromatography
amino acid analysis
SDS-PAGE
UV
IR and β-elimination reaction. Result: AmGP-2 was soluble in water with isoelectric point(pI) of 4.7 and relative molecular weight about 31 kDa
contents of proteins and carbohydrates were 56.7% and 29.3%
respectively; It contained 18 kinds of amino acids
higher content including aspartic acid
glutamic acid and lysine; Its carbohydrates chain portion contained arabinose
glucose and two kinds of undetermined monosaccharides. Conclusion: AmGP-2 had significant immunosuppressive activity
this study could provide a reference for development of immunodepressants of Chinese herbal medicines.
