Objective: To optimize ethanol extraction process of Bushen Huoxue granules. Method: HPLC was adopted to determine contents of cinnamaldehyde and amygdalin with mobile phase of acetonitrile(A)-water(B) for gradient elution(0-3.5 min
18%A;3.5-4.0 min
18%-55%A;4.0-9.0 min
55%A;9.0-9.5 min
55%-18%A)
detection wavelength at 290 and 210 nm
respectively.Taking transfer rates of cinnamaldehyde and amygdalin as a comprehensive evaluation index
effects of extracting method
soaking time and ethanol concentration on ethanol extraction process were investigated by single factor tests
orthogonal test was employed to optimize extraction process with ethanol consumption
extraction times and extracting time as factors. Result: Optimum extraction process was as follows:soaked 1 h with 10 times the amount of 60% ethanol
extracted thrice for 1.5 h each time;transfer rates of cinnamaldehyde and amygdalin were 85.43% and 86.95%
respectively. Conclusion: This optimized extraction technology was repeatable and feasible
it could provide a reference for industrial production of Bushen Huoxue granules.
