Objective: To study the effects of effective component groups of Qingfei Litan prescription on the acute lung injury in rats. Method: SD rats were randomly divided into nine groups:normal control group
model group
dexamethasone group
water extraction group
ethanol extraction group
petroleum ether extraction group
ethyl ether extraction group
n-butanol extraction group
residual water layer group. Acute lung injury model of rats was induced by intratracheal injection of lipopolysaccharide(LPS).The general situation of the rats was observed. The enzyme-linked immunosorbent assay (ELISA) was used to detect interleukin(IL) -1β
tumor necrosis factor alpha(TNF-α)
interleukint(IL-6) levels in serum. The pathological changes of lung tissue was observed in light microscopy. Nuclear factor-kappa B (NF-κB) p65 protein expressions were measured by immunohistochemistry in lung tissue. Result: Compared with normal control group
the lung tissue showed typical pathological features of inflammation and damage evidently in model group. Lung wet/dry weight ratio
the levels of IL-1β
TNF-α
IL-6 in serum
and the expressions of NF-κB p65 protein was significantly higher (P<0.01). Compared with the model group
effective component group of Qingfei Litan prescription and dexamethasone group show different degrees of mitigate trend in lung tissue damage
lung wet/dry weight ratio
the levels of IL-1β
TNF-α
IL-6 in serum
and the expressions of NF-κB p65 protein showed different degrees of mitigate trend
than water extraction group and ethanol extraction group had the most significant difference (P<0.01 or P<0.05)
and there was no significant difference compare with the dexamethasone group. Conclusion: The effective component groups of Qingfei Litan prescription can reduce pathological damage in the lung injury rats by LPS-induced. The mechanism may inhibit NF-κB protein signal transduction pathways
thereby reducing the secretion of cytokines (such as IL-1β
TNF-α
IL-6). The water extraction group and ethanol extraction group effect more obviously.
