Objective: To optimize extraction technology of Hirudo Zhusan. Method: The content of antithrombin activity substance was determined by anti-thrombin titration.Taking grinding degree as index to screen smash equipment
then effect of water content on powder of Hirudo Zhusan was investigated;with extract yield and transfer rate of antithrombin activity substance as indexes
single factor tests were adopted to optimize extraction process by taking soaking time
the amount of water
decoction time and so on as factors
then compared with traditional pieces of Hirudo. Result: Optimum extraction parameters was as follows:soaked 10 min with 20 times the amount of water
decocted thrice at 100 ℃ for 20 min each time;transfer rate of antithrombin activity substance could reach as highly as 93.31%
which was significantly higher than traditional pieces of 52.44%. Conclusion: This optimized process was stable and feasible
Hirudo Zhusan had advantages of high efficiency
short time
low energy consumption
environmental protection and so on
it could provide an insight for industrial production and new drug development of Hirudo Zhusan.
