目的:研究坐珠达西对N-甲基-N'-硝基-N-亚硝基胍(MNNG)致大鼠慢性萎缩性胃炎(CAG)的治疗作用。方法:大鼠自 由饮用170 mg ·L-1 的MNNG 8周建立大鼠CAG模型,之后随机分为模型对照组(10 mL ·kg-1 ·d-1)、摩罗丹组(0.36 g ·kg-1 ·d-1)、 坐珠达西高、中、低剂量组(每天ig 0.4,0.2,0.1 g ·kg-1 ·d-1)。给药9周后取材,观察大鼠胃的病理组织学改变,并对血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、白细胞介素12(IL-12)、一氧化氮(NO)进行检测。结果:与正常组比较,模型组大鼠胃部组织HE切片可见胃黏膜有炎性细胞浸润、充血和水肿等病理改变,血清中IL-12含量极显著降低(P<0.01),NO和MDA无显著性变化。与模型组比较,HE切片显示坐珠达西组能明显改善慢性萎缩性胃炎大鼠的胃黏膜病变,血清中NO含量显著降低(P<0.05),IL-12含量显著升高(P<0.01),MDA含量显著下降。结论:坐珠达西对MNNG所致萎缩性胃炎模型大鼠具有一定的治疗作用,机制可能与抑制炎症因子相关。
Abstract
Objective: To evaluate the effects of Zuozhu Daxi on rats with chronic atrophic gastritis (CAG)caused by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG). Method: CAG model was established by drinking 170 mg·L-1 MNNG for 8 weeks. Then the model rats were randomly divided into 5 groups
which was administered with distilled water(10 mL·kg-1·d-1)
Moluodan(0.36 g·kg-1·d-1)
Zuozhu Daxi (0.4
0.2
0.1 g·kg-1·d-1). At the end
the pathologic changes of gastric mucosa were observed
and malondialdehyde (MDA)
superoxide dismutase(SOD)
interleukin 12(IL-12)
nitric oxide(NO)in serum were tested. Result: Compared with the control group
the IL-12 of model group were decreased significantly(P<0.01).Compared with the model group
IL-12 levels in large and middle doses of Zuozhu Daxi were increased significantly (P<0.05
P<0.01)
the amount of NO and MDA was reduced significantly(P<0.05
P<0.01). SOD had no significant difference between groups. The stomach tissue sections stained by HE showed that the relief of pathological changes of gastric mucosa in the treatment groups were much better than model group. Conclusion: Zuozhu Daxi has therapeutic effect on rats with chronic atrophic gastritis caused by MNNG.
