Objective:The polysaccharide was separated and purified from Oldenlandia diffusa and the immuno-enhancing activity was evaluated in immunosuppression mice by intraperitoneal administration. Method: The crude polysaccharides were separated and purified by tangential-flow ultrafiltration
then the relative molecular weight was measured by gel column chromatography. The structure was analyzed by pre-column derivatization HPLC. The activity of the homogeneous polysaccharide was investigated by carbon clearance on immunosuppression mice. Result: A new homogeneous polysaccharide named ODP-1 was obtained from O. diffusa. The relative molecular weight of ODP-1 was about 20.88 kDa. ODP-1 was composed of mannose
rhamnose
galacturonic acid
glucose
galactose and arabinose with the molar ratio of 0.005: 0.033: 0.575: 1.000: 0.144: 0.143.Immune activity indicated that ODP-1 can improve the clearance index
phagocytic index
the index of thymus and spleen of immunosuppression mice. Conclusion: ODP-1 from O. diffusa is a homogeneous polysaccharide with immuno-enhancing activity.
