目的: 观察柴胡疏肝汤对慢性颞叶癫痫-抑郁共病模型大鼠海马中5-羟色胺(5-HT)含量及白介素-1β(IL-1β)
白介素-6(IL-6)mRNA表达的影响。方法: 腹腔注射氯化锂-匹罗卡品制作癫痫-抑郁共病大鼠模型
将成功模型随机分为模型组
西酞普兰组
柴胡疏肝汤高、中、低剂量组。正常组10只。药物干预组分别给予西酞普兰10 mg ·kg-1 ·d-1
柴胡疏肝汤高、中、低剂量10

2.5 g ·kg-1 ·d-1
正常组与模型组对应给予等量生理盐水ig
连续4周
2次/d。通过强迫游泳(FST)和蔗糖消耗试验评估抑郁行为的变化;视频录像监测大鼠痫性发作次数;液相色谱-质谱法检测海马5-HT的含量;实时荧光定量PCR检测海马IL-1β
IL-6 mRNA的表达量。结果: 与正常组比较
模型组大鼠FST累计不动时间明显延长
IL-1β mRNA表达量明显增加
糖水消耗量明显减少(P<0.01)
5-HT含量减少
IL-6 mRNA表达量增加(P<0.05)。与模型组比较
药物干预4周后
西酞普兰
柴胡疏肝汤高、中剂量组大鼠痫性发作次数明显减少
FST累计不动时间明显缩短
糖水消耗量明显增加
海马中IL-1β
IL-6 mRNA的表达明显下调(P<0.01﹚;5-HT的含量增加(P<0.05﹚。西酞普兰组与柴胡疏肝汤高、中剂量组比较
各指标差异均无统计学意义。结论: 柴胡疏肝汤高、中剂量具有抑制慢性颞叶癫痫-抑郁共病模型大鼠海马中IL-1β
IL-6 mRNA表达
促进5-HT表达
减少大鼠痫性发作次数
改善抑郁行为的作用。
