目的: 观察参芪复方对2型糖尿病大血管病变小鼠糖脂代谢的影响和对骨骼肌的保护作用。 方法: 自发性2型糖尿病KKAy小鼠60只
实验分为KKAy组、模型组、参芪复方组(14.4 mg ·g-1 ·d-1)和罗格列酮组(1.33 μg ·g-1 ·d-1)
每组15只
另设C57BL/6J小鼠15只为正常组
共5组。除正常组和KKAy组外
其余各组每日给予含一氧化氮合酶(NOS)抑制剂N-硝基-L-精氨酸甲酯(L-NAME) 的饮水及高脂饲料
连续8周
建立2型糖尿病大血管病变模型。造模同时开始ig给药
期间观察动物一般状况
监测各个时间段的血糖值
实验结束后摘眼球取血
分离血清
检测总胆固醇(TC)
甘油三酯(TG)
高密度脂蛋白胆固醇(HDL-C) 及低密度脂蛋白胆固醇(LDL-C)含量
取主动脉和腓肠肌并观察其病理形态学改变。 结果: 与正常组比较
KKAy组小鼠摄食量、饮水量、尿量偏多
体重较重
模型组与KKAy组小鼠血糖明显升高(P<0.01)
KKAy组小鼠血糖略高于模型组
但无差异
模型组小鼠血清中TC
TG
HDL-C
LDL-C含量均明显升高(P<0.01)
KKAy组小鼠血清中TC
TG
HDL-C
LDL-C含量均有所升高(P<0.05)
但低于模型组小鼠
模型组腹主动脉内膜水肿
出现明显的骨骼肌细胞萎缩、水肿、断裂和炎症状况
KKAy组小鼠均也有同模型组的变化
但无模型组明显
模型组与KKAy组小鼠在体表均可见到不同程度的抓痕及伤口;与模型组比较
参芪复方可改善模型动物的一般状态
参芪复方与罗格列酮均明显降低小鼠血糖(P<0.05
P<0.01)
均明显降低TC
TG
HDL-C
LDL-C含量(P<0.05
P<0.01);病理检查结果显示
参芪复方与罗格列酮均可减轻模型小鼠腹主动脉内膜水肿
减轻骨骼肌细胞萎缩、水肿、断裂和炎症状况。 结论: 参芪复方可改善2型糖尿病大血管病变小鼠糖脂代谢和血管损害
对骨骼肌起保护作用。
Objective: To observe the effect of Shenqi compound recipe on the glucose and lipid metabolism and the protective effect on skeletal muscle of mice with type 2 diabetic macroangiopathy. Method: Selected 60 KKAy mice were randomly divided into the KKAy group
the model group
the Shenqi compound recipe group (14.4 mg · g-1 · d-1) and the rosiglitazone group (1.33 μg · g-1 · d-1) (n=15)
another 15 C57BL/6J mice were assigned to the normal group. Spontaneous type 2 diabetic KKAy mice were induced by feeding water containing nitric oxide synthase inhibitor L-arginine methyl ester (L-NAME) and a high fat diet for 8 weeks except for mice in the KKAy group and the normal group. The general conditions
as well as blood sugar levels of animals were monitored. The levels of total cholesterol (TC)
triglyceride (TG)
high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterin (LDL-C) were detected. The morphological changes of the abdominal aorta and skeletal muscle were analyzed. Result: Compared with the normal group
the food intake
the water intake
the urine volume
the weight and the blood sugar increased in the KKAy group
the scratches and wounds were found in the body surface of the model and KKAy groups
the levels of TC
TG
HDL-C and LDL-C raised in the model (P<0.01) and the KKAy (P<0.05) groups. Compared with the model group

HDL-C and LDL-C declined significantly in the Shenqi compound recipe group (P<0.05

inflammatory state situation when compared with the model group. Conclusion: Shenqi compound recipe could improve the glucose and lipid metabolism
the vascular lesion of mice with type 2 diabetic macroangiopathy
and protect skeletal muscle.
