Objective: A method for rapid qualitative analysis of chemical constituents of Codonopsis pilosula by HPLC-LTQ-Orbitrap mass spectrometry was established. Mass information for different types of constituents was preliminarily approached. Method: A Phenomenex Kinetix C18(2.1 mm×100 mm
1.7 μm) column and a gradient elution of MeCN-H2O (0.01% formic acid) were used for HPLC separation. The combination of Orbitrap mass analyzer with a linear ion trap was applied for high resolution mass spectrometry and collision-induced dissociation (CID). Result: HPLC provided high separation efficiency of chemical components of C. pilosula. Based on the high-resolution mass information and MS/MS fragmentation behaviors of chemical components
19 compounds were identified or tentatively characterized. Conclusion: This work provided a good example for rapid analysis of different types of chemical constituents from C. pilosula. It might be helpful for further phytochemical studies and quality control of C. pilosula.
