摘要

目的: 对胡黄连苷-Ⅱ原料药进行结构鉴定

建立HPLC含量测定方法。方法: 根据理化常数和光谱数据进行结构鉴定;采用反相高效液相色谱法

对胡黄连苷-Ⅱ原料药进行含量测定。结果: 胡黄连苷-Ⅱ原料药结构鉴定表明其成分为6-香草酰基梓醇

即胡黄连苷-Ⅱ;HPLC测定胡黄连苷-Ⅱ在0.315~5.05 μg进样量与峰面积线性关系良好

平均加样回收率为98.09%

RSD 1.6%

3批胡黄连苷-Ⅱ原料药质量分数分别为101.53%

101.50%

102.25%。结论: 胡黄连苷-Ⅱ原料药含量较高

所建立含量测定方法简便可行

重复性好

可用于胡黄连苷-Ⅱ原料药的质量控制。

Abstract

Objective: To identify the structure of picroside-Ⅱ and establish an HPLC method for determining the content of picroside-Ⅱ. Method: The structure was elucidated by its physiochemical characteristics and spectral analysis. The content of picroside-Ⅱwas determined by HPLC. Result: The structure was elucidated as 6-vani11oyl catalpol (picroside-Ⅱ). The linear range of picroside-Ⅱwas within 0.315-5.05 μg. The average recovery was 98.09%

with RSD 1.6%. The three batches of active pharmaceutical ingredient picroside-Ⅱ were 101.53%

and 102.25%

respectively. Conclusion: The method is simple and specific

and can be used for control of picroside-Ⅱ.
