Objective: To establish an HPLC method for simultaneously determining eight ginsenosides and analyze their distribution in entire plant of Panax ginseng. Method: Discovery C18 column (4.6 mm×250 mm
5 μm) was adopted with the mobile phase of acetonitrile (A) and water (B) by the gradient elution. The flow rate was 1 mL·min-1
and the detection wavelength was at 203 nm. The RP-HPLC method was established for simultaneously determining Rg1

xylem of six-year-old ginseng plants. Result: The established method had a good practicable reproducibility and solution stability and separated eight components beyond the baseline
with a wider linear range and good linear relationship (r>0.999). The recoveries ranged from 98% to 102%. Conclusion: The distribution of eight ginsenosides in entire plant of ginseng was studied for the first time. The established method can be applied to simultaneously determine eight ginsenosides
with the maximum content in fibrous roots and buds and the minimum content in stems. Ginsenosides of main roots are mainly distributed in the cork layer.
