Objective: To study the effect of Curcumae Rhizoma oil on human osteosarcoma cell line saos-2
and discuss the change of the expression level of insulin-like growth factor-1 receptor (IGF-1R)
protein kinase B (Akt) and B-cell lymphoma/leukemia-2 (Bcl-2). Method: The effects of Curcumae Rhizoma oil on the proliferation of osteosarcoma cell line saos-2 were detected by MTT assay. Cytometry was used to analyze cell apoptosis; expression of IGF-1R was detected by real-time PCR
and the expression of Akt and Bcl-2 was detected by western-blot. Result: Curcumae Rhizoma oil could obviously inhibit the growth of osteosarcoma cells and induce saos-2 cell apoptosis in a dose dependent manner. The expression of IGF-1R
Akt and Bcl-2 was down regulated in the curcumol-treated cells. Conclusion: Curcumae Rhizoma oil can inhibit proliferation and induce apoptosis of saos-2 cells
and its antitumor effect is associated with suppressing the expression of IGF-1R
