目的:从赣南藤梨根中提取分离得到乌苏烷三萜化合物A(2β
3β
23-三羟基-12-烯-28-乌苏酸
ursolic compound A)
研究其对人宫颈癌HeLa细胞株的增殖抑制作用。方法:应用E-MEM培养基建立宫颈癌HeLa细胞体外培养体系
将4种不同浓度(0.5
1.0
2.0
4.0μmol·L-1)的ursolic compound A作用后的宫颈癌细胞(每孔含2000细胞)在96孔细胞培养板上培养24
48 h;细胞活力采用细胞计数试剂盒(CCK-8)检测
乳酸脱氢酶(LDH)活性用噻唑蓝(MTT)法测定
肿瘤细胞增殖情况采用免疫比色法检测。结果:与对照组相比
用0.5~4.0μmol·L-1 ursolic compound A处理过的HeLa细胞
其乳酸脱氢酶释放率随着剂量增大和时间延长而增加(P<0.05
P<0.01);CCK-8和5-溴脱氧尿嘧啶核苷(BrdU)检测结果表明
化合物A比对照药物抗宫颈癌活性相对较强
作用效果呈浓度与时间依赖性
宫颈癌细胞的活性也相应不断下降
同时该化合物对宫颈癌细胞株的增殖抑制作用也较明显(P<0.05
P<0.01)。结论:Ursolic compound A对HeLa细胞增殖有一定抑制作用。
Objective: To investigate the effect of 2β
3β
23-trihydroxy-urs-12-en-28-olic acid(ursolic compound A) from the root of Actinidia chinensis on the proliferation of cervical cancer HeLa cells. Method: HeLa cells(2×103 cells/well) treated by ursolic compound A of different concentrations(0.5

4.0μmol·L-1) were cultured in 96-well plates for 24
48 h after the cervical cancer HeLa cell lines in vitro culture system was established by Essential Medium-Eagle(E-MEM) medium;the viability of HeLa cells was detected by cell counting kit-8(CCK-8).The activity of lactic dehydrogenase(LDH) was measured by methyl thiazolyltetrazolium(MTT)assay. Immune colorimetry was used to test the proliferation of HeLa cells. Result: Compared with the control group
for HeLa cells treated by ursolic compound A of different concentrations
the release rate of LDH was increased in a dosage and time dependent manner(P<0.05
P<0.01).Results of BrdU and CCK-8 showed that
the inhibitory effect of ursolic compound A to HeLa cell was stronger than control drugs
and its inhibitory effect was in a dosage and time dependent manner. The activity of cervical cancer cells would be decreased accordingly
and at the same time
the compound had significant inhibition effect on the proliferation of cervical cancer cells(P<0.05
P<0.01). Conclusion: Ursolic compound A has an inhibitory effect on the proliferation of HeLa cells.
