目的:探讨四神丸对结肠炎大鼠派伊尔结(Payer's patch
PP结)中Treg/Th17细胞间平衡的影响。方法:将40只SD大鼠随机分为4组
分别为正常组、模型组、四神丸组(2.5 g·kg-1)及美沙拉嗪组(0.15 g·kg-1)。除正常组外
其余各组建立2

6-三硝基苯磺酸(TNBS)/乙醇法诱导的大鼠结肠炎模型后
ig给药7 d。采用流式细胞仪检测大鼠小肠PP结中T淋巴细胞亚群的表达水平并计算Treg/Th17
Western blot检测结肠组织中维甲酸相关孤核受体γt(RORγt)和信号转导和转录激活因子5a(STAT5a)的表达量。结果:与正常组比较
模型组CD4+T淋巴细胞水平明显升高
CD8+T细胞水平显著降低
且CD4/CD8间比例明显升高
CD4+CD25+Foxp3+Treg细胞明显降低
Th17细胞明显升高
Treg/Th17降低
而且结肠组织中RORγt蛋白表达量升高
STAT5a蛋白表达量则明显降低(P<0.05
P<0.01);与模型组比较
四神丸组大鼠小肠PP结中CD4+T淋巴细胞水平明显降低
CD8+T细胞水平显著升高
且CD4/CD8间比例明显降低(P<0.01)
同时
四神丸组则CD4+CD25+Foxp3+Treg细胞显著升高
Th17细胞明显降低
Treg/Th17升高
而且结肠组织中RORγt蛋白表达量降低
STAT5a蛋白表达量则显著增加(P<0.05
P<0.01)。结论:四神丸可以调节T淋巴细胞亚群
并通过抑制RORγt表达、提高STAT5a的蛋白分泌调节Treg/Th17间的平衡。
Objective: To investigate the effect of Sishen Wan on the balance of Treg/Th17 in Payer's patch of colitis rats. Method: The forty SD rats were randomly divided into 4 groups:normal group
model group
Sishen Wan group (2.5 g·kg-1) and mesalazine group (0.15 g·kg-1). Except the normal group
all other groups established the rat colitis models induced by 2

6-trinitrobenzene sulfonic acid (TNBS). Drugs were given intragastrically for 7 days. The levels of T lymphocyte subsets were measured by flow cytometry
and the ratio of Treg/Th17 was then calculated. The expressions of retinoid acid-related orphan receptor γt (ROR-γt) and signal transducer and activator of transcription 5a (STAT5a) were tested by the Western blot. Result: Compared with the normal group
the model group significantly increased the levels of CD4+T lymphocyte
significantly decreased CD8+T cells
significantly increased the ratio of CD4/CD8
significantly decreased CD4+CD25+Foxp3+Treg cells
significantly increased Th17 cells
decreased the ratio of Treg/Th17
increased the expressions of ROR-γt protein in colon tissues
and significantly decreased the expressions of STAT5a protein (P<0.05
P<0.01). Compared with the model group
Sishen Wan group significantly decreased CD4+T lymphocyte in Payer's patch of rats
significantly increased CD8+T cell levels
and significantly decreased the ratio of CD4/CD8 (P<0.01). At the same time
Sishen Wan group significantly increased CD4+CD25+Foxp3+Treg cells
decreased Th17 cells
increased the ratio of Treg/Th17
decreased the expressions of ROR-γt protein in colon tissues
and significantly increased the expressions of STAT5a protein (P<0.05
P<0.01). Conclusion: The Sishen Wan could regulate the T lymphocyte subsets
and by inhibiting the expressions of RORγt and increasing the protein secretion of STAT5a
it can also regulate the balance of Treg/Th17.
