原位末端标记法(TUNEL)检测细胞凋亡率;激光共聚焦显微镜测定线粒体膜电位(MMP)和钙离子(Ca2+)荧光强度;逆转录-聚合酶链式反应(RT-PCR)检测mRNA表达;蛋白质免疫印迹(Western blot)检测磷脂酰肌醇3激酶-蛋白激酶(PI3K-Akt)表达。 结果: 与空白组比较
模型组GC凋亡率显著增加
MMP下降
Ca2+荧光强度升高
细胞死亡的B淋巴细胞瘤-2基因抑制剂(Bad)
半胱氨酸蛋白酶-3(Caspase-3)
Caspase-9 mRNA表达均显著上调
B淋巴细胞瘤-2(Bcl-2) mRNA则显著下降;PI3K
p-Akt蛋白表达显著下调(P<0.05);中、高剂量组MMP显著升高

31.5 g·kg-1) were used to intervene in apoptosis. The blank group was also set up. The proliferation and apoptosis were analyzed by MTT assays and flowcytometry. down-regulating expressions of pro-apoptosis factors (Bad and caspases family)
stabilizing the mitochondrial membrane potential and reducing calcium influx.
