Objective: To extract the volatile oil and small polar components of the Hui formula Muxiang oil by steam distillation (SD) and supercritical CO2fluid extraction (SFE)
respectively;analyze and identify the components and their contents with Gas Chromatography-Mass Spectromet (GC-MS)
and compare the differences in types of components between different extraction methods. Method: The mass spectrometric conditions were set as follows:Shimadzu QP2010 plus GC-MS
Rxi-5Sil MS quartz capillary column (0.25 mm×30 m
0.25 μm)
the starting temperature of 50℃
maintained for 1 min
heated to 150℃ with 8℃·min-1
maintained for 3 min
then heated to 210℃ with 3℃·min-1
maintained for 3 min
and then heated to 280℃ with 15℃·min-1
maintained for 15 min until completion of analysis;helium as the carrier gas
column flow rate at 1.33 mL·min-1
split ratio of 25:1
inlet temperature of 250℃
EI ionization source of 70 eV
ion source temperature of 230℃
and scan range of m/z 35-500. Result: Yield of volatile oil was 0.8% and 2.91% respectively in SD and SFE methods;25 compounds were identified
accounting for 96.28% of the total peak area of volatile oil in SD method;36 compounds were identified
accounting for 97.98% of the total peak area of volatile oil in SFE method. Conclusion: There was significant difference in types of components contained in the volatile oil between SD and SFE methods
and GC-MS could be used to identify the small polar components of the medicine
providing experimental basis for further development and utilization of Hui medicine.
