目的: 研究健骨片对去卵巢骨质疏松大鼠骨代谢、骨质微结构及骨强度的作用。方法: 将50只雌性SD大鼠随机分为假手术组(n=10)
模型组(n=10)
戊酸雌二醇片组(n=10)
健骨片低剂量组(n=9)
健骨片高剂量组(n=9)。除假手术组外
其余各组行卵巢切除建立大鼠骨质疏松模型
术后各组分别灌服相应药物或蒸馏水
给药剂量分别为戊酸雌二醇片组(0.1 mg·kg-1·d-1)
健骨片低剂量组(0.35 g·kg-1·d-1)
健骨片高剂量组(1.05 g·kg-1·d-1)
连续12周。每周测定大鼠体重
给药结束后酶联免疫吸附测定(ELISA)测定大鼠血清骨钙素(OC)。测定血清碱性磷酸酶(ALP)
抗酒石酸酸性磷酸酶(StrACP)
钙(Ca)
磷(P)。Micro-CT扫描重建股骨微结构
检测骨矿含量(BMC)
组织骨矿含量(TMC)
骨密度(BMD)
组织矿物质密度(TMD)
骨体积分数(BV/TV)
骨小梁数量(TbN)
骨小梁分离度(TbSp)
骨小梁厚度(TbTh)。三点弯曲法测定股骨刚度及最大载荷。结果: 与假手术组比较
模型组大鼠血清OC
ALP显著升高
血清Ca
P明显降低(P<0.05
P<0.01);股骨远心端及中点骨小梁稀疏
缺失严重
BMC
TMC
BMD
TMD
BV/TV明显降低(P<0.05
P<0.01);股骨最大载荷显著降低(P<0.05)。与模型组比较
健骨片低、高剂量组均明显降低血清OC及ALP
升高血清Ca

P<0.01)。结论: 健骨片可影响去卵巢骨质疏松大鼠骨代谢
从而改善骨微结构
增加骨强度。
Objective: To investigate the effect of Jiangu tablets on bone metabolism
microarchitecture and stiffiness in an ovariectomized rat model. Method: Totally 50 Sprague-Dawley rats were randomly divided into the sham group (n=10)
the model group (n=10)
the estradiol valerate tablets group (n=10)
the low-dose Jiangu tablets group (n=9) and the high-dose Jiangu tablets group (n=9). Ovaries were removed by surgery in each group to establish an ovariectomized rat model except sham group. Different groups were treated with different drugs or water. Estradiol valerate tablets group was administrated with estradiol valerate tablets at 0.1 mg·kg-1·d-1
low-dose Jiangu tablets group was administrated with Jiangu tablets at 0.35 g·kg-1·d-1
high-dose Jiangu tablets group was administrated with Jiangu tablets at 1.05 g·kg-1·d-1. The treatment maintained for 12 weeks. The body weight of rats was detected every week. Serum concentration of osteocalcin (OC)
alkaline phosphatase (ALP)
tartrate-resistant acid phosphatase (StrACP)
calcium (Ca)
and phosphorus (P) were determined after treated. Bone microarchitecture and bone mineral content (BMC)
tissue mineral content (TMC)
bone mineral density (BMD)
tissue mineral density (TMD)
bone volume fraction(BV/TV)
trabecula number (TbN)
trabecular separation (TbSp)
trabecular thickness (TbTh) were detected by Micro-CT. The femur maximum load and stiffness were detected by a three-point bending testing. Result: Compared with the sham group
the levels of OC and ALP were increased significantly
the levels of Ca and P were decreased significantly (P<0.05
P<0.01)
the trabecula of distal and middle femur in model group became rare

and the level of TbSp was significantly decreased in the high-dose Jiangu tablets group(P<0.05). The maximum load and stiffness of femur were significantly increased in the low-and high-dose Jiangu tablets group (P<0.05
P<0.01). Conclusion: Jiangu tablets influenced the bone metabolism
ameliorated the bone microarchitecture and increased the bone stiffness in an ovariectomized rat model.
