目的: 研究桑椹提取物对四氯化碳(CCl4)及乙醇致小鼠急性肝损伤的保护作用。 方法: 分别取昆明种小鼠60只,雌雄各半,随机分为6组,分别为正常组和模型组(ig给予蒸馏水),阳性药(联苯双酯滴丸)组(0.40 g·kg-1),桑椹提取物低、中、高剂量组(1.525,3.050,6.100 g·kg-1),除正常组外分别采用CCl4和乙醇建立小鼠急性肝损伤模型,造模成功后各组连续ig给药7 d。给药后测定小鼠肝脏系数,测定CCl4肝损伤小鼠血清中丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST)活性,以及酒精性肝损伤小鼠肝组织匀浆中甘油三酯(TG),还原型谷胱甘肽(GSH)和丙二醛(MDA)的含量,苏木素-伊红(HE)染色观察肝组织病理切片。 结果: 与正常组比较,模型组CCl4肝损伤小鼠血清中ALT,AST活性明显升高(P<0.01),酒精性肝损伤小鼠肝组织中MDA和TG含量明显升高,GSH含量明显降低(P<0.01),模型组病理学观察肝损伤均较为明显;与模型组比较,桑椹提取物能降低ALT,AST活性,能明显降低MDA和TG含量,明显升高GSH含量,均具有明显的统计学差异(P<0.05,P<0.01),病理检查结果显示玉热买提取物有明显的保肝作用。 结论: 桑椹提取物对小鼠四氯化碳、酒精肝损伤有良好的保护作用。
Objective: To observe the protection effect of extract from Xinjiang mulberry on carbon tetrachloride (CCl4) and alcohol-induced acute liver injury in mice. Method: Totally 60 Kunming mice (30 males and 30 females) were randomly divided into 6 groups:normal group and model group with intragastric administration of distilled water
positive group (bifendate pills

6.100 g·kg-1). All the other mice except those in normal group received CCl4 and alcohol to establish acute liver injury models in mice. After successful modeling
the mice in all groups received intragastric administration for continuous 7 days. The liver coefficient in mice was determined after drug administration
and colorimetric method was used to determine the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum of CCl4 liver injury mice
and the contents of triglyceride (TG)
malondiadehyde (MDA) and glutathione (GSH) in liver homogenate of alcoholic liver injury mice. HE staining method was used to observe the pathological section of liver tissues in mice. Result: As compared with the normal group
the activities of ALT and AST in serum of CCl4 liver injury mice in model group were significantly increased (P<0.01);the contents of MDA and TG in liver tissues of alcoholic liver injury mice in model group were significantly increased while the content of GSH was significantly reduced (P<0.05
P<0.01);the liver injury was more obvious in the model group during the pathological observation. As compared with the model group
the extract from Xinjiang mulberry could reduce the activities of ALT and AST in serum
significantly reduce the contents of TG and MDA
and significantly increase the content of GSH
with statistically significant differences (P<0.05
P<0.01). The pathological examination results indicated that the extract from Xinjiang mulberry had obvious liver-protecting effect. Conclusion: The extract from Xinjiang mulberry has notable protective effect for the CCl4 and alcohol-induced liver injury in mice.
