Objective: To establish an HPLC method for the simultaneous determination of four flavonoids components in flowers of Coreopsis tinctoria. Method: The chromatographic separation was performed on Cosmosil-5C18-AR-Ⅱ column (4.6 mm×250 mm
5 μm) with acetonitrile-0.1% acetic acid solution as the mobile phase for gradient elution
at a flow rate of 1.0 mL·min-1
and the detection wavelength was set at 280 nm (for flavanomarein
isookanin) and 378 nm (for marein
okanin) to determine the contents of flavanomarein
isookanin
marein and okanin in Coreopsis tinctoria. Result: 4 flavonoids components were well separated
with good linear relationship between 0.503 5-5.035 μg

and average recovery rate was between 95.4%-98.4%. Conclusion: The method is accurate and selective
which could provide reference for the quality control of C. tinctoria.
