Objective: To clarify the pathogenic species of Gardenia jasminoides brown spot
and provide the basis for disease prevention and control. Method: The diseased leaf with G. jasminoides brown spot were collected from different regions of Jiangxi province. Pathogens were separated and purified by tissue culture method
and identified by using pathogenic test
pathological observation and rDNA-ITS sequence analysis according the Koch's postulates. Pathogenicity of the leaf from different species plants was identified by using isolated leaf inoculation method to determine the host range. Result: The pathogens separated from sample leaves showed white villiform aerial hyphae at the initial stage in medium
so that the color of colony was white at first
and then turned to dark brown. Observation under microscope showed that the hyphae had septum and branch
with gray to brown spheroidal fruiting body. The separated pathogen would make the healthy G. jasminoides leaves produce brown spot after tie-back. Blast analysis of the amplificated rDNA-ITS sequence confirmed that the disease was caused by Botryosphaeria dothidea. The pathogen had a wide host range
and the test results of 10 kinds of plants showed strongest pathogenicity in Peucedanum praeruptorum
but showed no pathogenicity in Gomphrena globosa. Conclusion: The pathogens and host range for G. jasminoides brown spot were clarified in this paper.
