目的:观察加减四妙散对动脉粥样硬化大鼠血脂、降钙素、抵抗素及主动脉弓部病理状态的影响。方法: SPF级SD大鼠60只雌雄各半,随机分为6组,分别为正常组,阳性药组(阿托伐他汀,0.83 mg·kg-1),模型组,加减四妙散低、中、高剂量组(0.325,0.650,1.300 g·kg-1)。除正常组外,余以18%猪油,7%胆固醇,1%牛胆盐与花生油配成糊状乳糜液,按照10 mL·kg-1每天ig 1次,连续8周造模,8周后给药,同时维持造模,给药6周后禁食12 h,处死取血液检测总胆固醇(TC),甘油三酯(TG),血糖(Glu),低密度脂蛋白胆固醇(LDL-C),高密度脂蛋白胆固醇(HDL-C),降钙素基因相关肽(CGRP)及抵抗素含量,计算大鼠肝脂数,Lees肝脂数及脂肪指数,取心脏及主动脉做病理切片,苏木素-伊红(HE)染色观察各组主动脉病理状况。结果:与正常组比较,模型组大鼠血清中TC,TG,Glu,LDL-C及抵抗素含量均升高,肝指数,Lees指数及脂肪指数均明显增加,HDL-C,CGRP含量明显降低(P < 0.05,P < 0.01),同时病理显示高脂模型大鼠出现主动脉硬化早期病变,证明实验成功复制了动脉粥样硬化早期模型;与模型组比较,阿托伐他汀组、加减四妙散高组明显降低TC,TG,Glu,LDL-C及抵抗素含量,加减四妙散中、高组明显降低肝指数,Lees指数及脂肪指数(P < 0.05,P < 0.01),加减四妙散中、高组明显升高HDL-C,CGRP含量(P < 0.05,P < 0.01),病理提示其病理发展较模型组有改善。结论:四妙散颗粒可以降低血脂,降低低密度脂蛋白,并通过调节炎症因子来改善动脉粥样硬化,减缓其发展,对动脉粥样硬化预防与治疗有积极作用。

the rats were fasted for 12 h before killed to detect blood content of total cholesterol (TC)

and calculate rat liver index
Lee's liver index and fat index. Their heart and aorta were collected to make pathological sections. HE staining was performed to observe aortic pathological conditions of each group. Result: Compared with the blank control group

pathology results showed early lesions of aortic atherosclerosis among hyperlipidemia rats. This proves that the experiment successfully replicated the early atherosclerosis model. Compared with the model group
the positive group and the modified Simiaosan high-dose group showed notable reduction in TC

LDL-C and resistin content
modified Simiaosan medium and high-dose groups showed notable reduction in rat liver index
Lee's liver index and fat index (P < 0.05
P < 0.01) and remarkable increase in HDL-C and CGRP content (P < 0.05
P < 0.01). The pathological results also showed alleviation in pathological process compared with the model group. Conclusion: Modified Simiaosan may reduce cholesterol
LDL and alliviate atherosclerosis by regulating inflammatory cytokines
with a good effect in atherosclerosis prevention and treatment.
