目的:观察不同配比甘草-芫花煎剂对肝癌腹水模型小鼠的干预影响,并探讨其作用机制。方法:取昆明种雄性小鼠135只,随机分为正常组、模型组、芫花单煎组、芫花-甘草(1:0.25,1:0.5,1:1,1:2,1:4)组和甘草单煎组。除正常组外,其他组小鼠腹腔均接种肝癌腹水瘤细胞系H22,并分别灌胃给药,正常组和模型组给予等容积生理盐水。12 d后分别测量各组小鼠的腹水量、腹围和体重及肝肾指数,蛋白质免疫印迹(Western blot)法测定肾脏组织中水通道蛋白2(AQP2)和肾加压素受体(V2R)的蛋白表达,生化法测定小鼠血清中丙氨酸氨基转移酶(ALT),天门冬氨酸氨基转移酶(AST),尿素氮(BUN)和尿肌酐(Cr)的水平。结果:与正常组比较,模型组小鼠的腹水量、腹围、体重、肝指数及血清中ALT,AST,BUN,Cr水平均显著升高(P<0.01),肾指数显著降低(P<0.01),AQP2,V2R蛋白的表达显著升高(P<0.01)。与模型组比较,芫花单煎组和芫花-甘草1:0.5,1:1和1:2组均能显著降低小鼠的腹水量和体重(P<0.05,P<0.01);芫花单煎组和芫花-甘草1:1组同时能显著降低小鼠的腹围(P<0.05,P<0.01),降低AQP2,V2R蛋白的表达(P<0.05,P<0.01)。与芫花单煎组比较,芫花-甘草1:1组能显著降低小鼠的腹水量、腹围和体重(P<0.05,P<0.01),降低AQP2,V2R的表达(P<0.05),芫花-甘草1:4组则使其显著升高(P<0.05,P<0.01)。结论:不同比例甘草的配伍能增强或抵消芫花抗小鼠肝癌腹水的作用,且相关变化可能与其有效调节了肾组织内的AQP2和V2R蛋白表达有关。

The underlying mechanism might be correlated with the effective regulation of aquaporin-2 and renal vasopressin receptors-2 protein expression.
