目的:研究交泰丸中黄连、肉桂不同配伍比例(10:1,10:3,10:6)对db/db小鼠的降糖作用及其相关机制。方法: 12周龄db/db糖尿病小鼠随机分为黄连-肉桂10:1组,10:3组,10:6组(相当于生药8.4,10,12.3 g·kg-1),阳性药物组(二甲双胍,0.25 g·kg-1),模型组,同月龄db/m小鼠作为正常。正常组和模型组以生理盐水灌胃,阳性药物组以二甲双胍混悬液灌胃,不同配伍比例交泰丸以相应中药水煎剂灌胃,每天1次。5周后,观察小鼠饮水量、进食量、体重的变化,检测空腹血糖(FBG),并进行腹腔注射葡萄糖耐量实验(IPGTT)和胰岛素耐量实验(IPITT);胰岛素免疫组化染色以及电子显微镜观察胰岛β细胞形态及超微结构的变化;蛋白质免疫印迹法(Western blot)分析肝脏、腹部脂肪与肌肉组织中的AMP依赖的蛋白激酶(AMPK)与磷酸化AMPK(p-AMPK)的表达,以及脂肪和肌肉组织中葡萄糖转运蛋白4(GLUT4)表达的变化。结果:与正常组比较,模型小鼠体重、饮水量、进食量增加(P<0.01),FBG显著升高(P<0.01),糖耐量与胰岛素耐量各时间点血糖显著升高(P<0.01),胰岛β细胞数量及细胞内分泌颗粒数量减少,肝脏、脂肪和肌肉组织中p-AMPK表达降低(P<0.05),脂肪和肌肉组织中GLUT4表达降低(P < 0.01,P < 0.05);与模型组比较,交泰丸3个不同配比组FBG均明显降低(P<0.05),IPGTT和IPITT明显改善,胰岛β细胞数量和分泌颗粒较模型组明显增多,其中以黄连-肉桂10:1组方的作用最为明显;交泰丸10:1组能明显增加肝脏、脂肪和肌肉中的p-AMPK水平(P<0.05),增加脂肪和肌肉组织中GLUT4的表达(P < 0.01,P < 0.05)。结论:黄连-肉桂10:1,10:3,10:6不同配比的交泰丸对db/db小鼠均具有一定降血糖作用,但以10:1比例效果最好,其作用机制与激活AMPK,增加GLUT4的表达有关。

the numbers of pancreas islet β cells and granules in β cells were higher than the model group. Among three groups
the effect of Rhizoma coptidis and cinnamon with the proportion of 10:1 was the most obvious. The expression of phosphorylated AMPK was increased in the liver
adipose tissue and muscle of db/db mice treated with Jiaotai pill 10:1 group (P<0.05). And the expression of GLUT4 was also increased in adipose tissue and muscle of Jiaotai pill 10:1 group (P < 0.05
P < 0.01). Conclusion: Jiaotai pill composed of Rhizoma coptidis and cinnamon with different proportions has a certain effect in decreasing blood glucose
and that with the proportion of 10:1 showed the best effect. Its mechanism is related to activation of AMPK and increase in the expression of GLUT4.
