Objective: To investigate the protective effect of Gualou Guizhi granule on oxidative stress and its molecular mechanism. Method: Magnetic resonance imaging (MRI) was performed to measure cerebral infarction area. The levels of reactive oxygen species (ROS)
malondialdehyde (MDA) activity and antioxidant enzyme activities of superoxide dismutase (SOD)
catalase (CAT) and glutathione peroxidase (GSH-Px) activity were determined by respective kits. Meanwhile
Western blot and Real-time PCR were used to detect the protein and gene expressions of NF-E2-related factor 2 (Nrf2)
heme oxygenase-1 (HO-1)
NAD(P)H: quinone oxidoreductase 1 (NQO1) and Kelch-like ECH-associated protein1 (Keap1). Result: Gualou Guizhi granule can significantly reduce the area of cerebral infarction and the production of MDA and ROS (P < 0.05)
and increase the activities of SOD
CAT and GSH-Px (P < 0.01). In addition
Western blot and Real-time PCR showed that Nrf2
HO-1
Keap1and NQO1 mRNA and protein expressions were up-regulated (P < 0.05) by Gualou Guizhi granule. Conclusion: Gualou Guizhi granule may inhibit oxidative stress by suppressing Nrf2 pathways in the rat model with cerebral ischemia-reperfusion injury.
