Objective:To investigate the effects of Cordyceps protein extract (OSPE) on the growth of A549 lung cancer cells and immune activity of mouse peritoneal macrophage in vitro. Method:A549 cells were treated with low

respectively. Cell viability was detected by methyl thiazolyl tetrazolium (MTT) method; cell apoptosis and necrosis were evaluated by Hoechst 33258 staining and flow cytometry. Real-time PCR was used to detect the mRNA expression of Bcl-2 associated X protein (Bax) and B cell leukemia 2 (Bcl-2).Enzyme linked immunosorbent assay (ELISA) was optimized to detect the effect of OSPE on macrophage secretion of tumor necrosis factor-α (TNF-α)
interleukin-1 (IL-1) and interleukin-12 (IL-12) levels. Result:The (100

500 mg·L-1) OSPE remarkably inhibited the A549 cell growth with an inhibitory rate of 19.29%

and 200 mg·L-1 OSPE (12-72 h) decreased significantly cell growth by increasing apoptosis in a time dependent manner. After Hoechst staining
the cells showed typical morphological changes of apoptosis
and the flow cytometry results showed that 200 mg·L-1 OSPE increased the apoptosis of A549 cells in a time dependent manner (P<0.01). Real-time PCR results showed that OSPE significantly increased Bax mRNA expression and significantly decreased Bcl-2/Bax expression (P<0.01). The results of ELISA showed that OSPE significantly promoted TNF-α
IL-1 and IL-12 levels in normal macrophages (P<0.01). Conclusion:OSPE inhibited the growth of A549 cells by inducing apoptosis
also probably by promoting the secretion of the macrophage secretion factors.
