Objective:To explore the genetic differentiationof Polygonum sibiricum in different areas in China
in order toprovide basis for its population subdivision
evolution
protection and utilization
and quality control. Method:Fiftysix samples were collected from 13 sampling sites in four provinces of China. EST-SSR primers were developed and screened
and the genetic differentiation was evaluated with both EST-SSR and sequence related amplified polymorphism(SRAP) markers. Result:Among 45 pairs of EST-SSR primers
38 pairs amplified 660 distinguished bands
and 17 pairs amplified 303 polymorphic bands
with a polymorphism rate of 100%. Among 96 pairs of SRAP primers
67 pairs amplified 1 347 distinguished bands
and 58 pairs amplified 1 147 polymorphic bands
with a polymorphism rate of 100%. Genetic distance and cluster analysis revealed both inter-and inner-population have genetic differentiation. Conclusion:Genetic variation and geographic differentiation areobserved among P. sibiricum populations in China
