目的:通过研究黔产莪术油对直肠癌SW1463细胞中死亡因子受体(Fas)的蛋白表达及分泌的免疫因子Toll样受体2(TLR2),Toll样受体4(TLR4),白细胞介素-10(IL-10),癌基因C-Raf转化生长因子-β1(TGF-β1)的影响,探讨黔产莪术油对肿瘤细胞的免疫效应。方法:将水蒸气蒸馏提取的黔产莪术油,配置成空白组,黔产莪术油80,120,160,200 mg ·L-1组,作用于直肠癌细胞SW1463细胞24 h后,采用免疫酶联免疫吸附测定(enzymeLinked immunosorbent assay,ELISA)检测不同质量浓度黔产莪术油对IL-10水平的影响,采用蛋白免疫印迹法(Western blot)检测Fas,TLR2,TLR4,C-Raf,TGF-β1蛋白表达对直肠癌细胞SW1463的增殖抑制作用。结果:ELISA检测发现,与空白组比较,莪术油组IL-10水平随着药物浓度的升高表达降低(F=23.11,P<0.01)。Western blot结果显示,与空白组比较,莪术油组随着药物浓度的升高,TLR2,TLR4,C-Raf蛋白表达显著降低(P<0.01);Fas,TGF-β1蛋白表达明显降低(P<0.05)。结论:黔产莪术油抑制SW1463细胞的作用免疫作用机制可能与下调Fas/FasL通路有关,使得Toll样受体TLR2,TLR4蛋白表达下调,导致相关因子TGF-β1的表达下调,从而使得细胞凋亡,起到免疫增强作用。

C-Raf and TGF-β1 protein expression levels and investigate its inhibitory effect on rectal cancer cells SW1463 proliferation. Result:ELISA results showed that as compared with blank control group
IL-10 expression was down-regulated with the increase of concentration in Guizhou zedoary oil groups (F=23.11
P<0.01). Western blot results showed that the protein expression levels of TLR2
TLR4 and C-Raf were significantly decreased with the increase of drug concentration (P<0.01)
and the protein expression levels of Fas and TGF-β1 were also decreased (P<0.05). Conclusion:The immune mechanism of Guizhou zedoary oil in inhibiting SW1463 cells may be associated with down-regulation of Fas/FasL pathway
down-regulation of Toll like receptor TLR2 and TLR4 protein expression
leading to the down-reulation of relate factor TGF-β1
so that cell apoptosis is achieved to enhance the immune function.
