Objective:To establish the methods of content determination of stachydrine hydrochloride in Leonurus granule by high performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD). Method:The samples were treated with 732 sodium cation exchange resin. The determination was performed on Venusil HILIC column(4.6 mm×250 mm
5 μm);column temperature:room temperature;with acetonitrile and 0.2% acetic solution as mobile phase (80:20) at the flow rate 1 mL·min-1;sample amount 10 μL;drift tube temperature 79℃;air volume flow 2 L·min-1. Result:The stachydrine hydrochlorideshowed a good linear relationship within the range of 0.498-4.980 μg(r=0.999 5
n=6);and the average recovery was 99.7% and 99.3% respectively for sugar free Leonurus granule and sugar containing Leonurus granule. Conclusion:The method is specific
accurate and simple
which can be applied as a new way for content determination of Leonurus granule
and provide reference for content determination of stachydrine hydrochloride in other unilateral preparations of Leonuri Herb.
