Objective:A great deal of researches have shown that Populus tomentosa male inflorescence
as our traditional Chinese medicine
has a variety of chemical constituents and strong anti-bacterial
anti-inflammatory and other pharmacological effect. The aim of this study was to investigate the chemical constituents of P. tomentosa systemically to enrich its research materials and lay foundations for the development of new drugs and its clinical application. Method:The male anthotaxy of P. tomentosa male inflorescence(30 kg)was extracted for three times with 70% ethanol
concentrated under reduced pressure
and then the extract was suspended in water. Then the solvent was partitioned with petroleum ether
EtOAc and n-BuOH successively. The components of EtOAc extract were isolated and purified by ODS
silica gel
Sephadex LH-20 column chromatography and recrystallization. Their structures were identified based on the physiochemical properties and spectroscopy techniques such as ESI-MS and nuclear magnetic resonance (NMR). Result:Twelve compounds were isolated from the ethyl acetate fraction
and they were identified as quercetin(1)
taxifolin(2)
luteolin(3)
eriodictyol(4)
myricetin(5)
dihydromyricetin(6)
(2S)-5
7-dihydroxyflavanone-7-O-β-D-glucopy-ranoside(7)
protocatechuic acid(8)
caffeic acid(9)
2-acetyl-1
3-dicaffeoylglycerol(10)
salicortin-6'-benzoate(11)
and daucosterol(12)
respectively. Conclusion:Compounds 2
5-7 were isolated from the Populus genus for the first time
and Compounds 2
4-8
10
11 were isolated from the plant for the first time
providing certain chemical basis for comprehensive utilization of Populus tomentosa.
