目的:

探究使药石菖蒲对定志小丸中人参皂苷成分肠吸收的影响,从肠吸收角度揭示石菖蒲“佐使则有功”的科学内涵。
方法:

采用超高效液相色谱-串联质谱法(UPLC-MS/MS)测定大鼠样品中人参皂苷Rg
1

的浓度;通过大鼠在体肠灌流试验比较不同配伍方式下定志小丸中3种人参皂苷类成分肠吸收速率常数(
K
a
)与表观通透系数(
P
app
)的差异;通过大鼠外翻肠囊试验考察不同剂量石菖蒲挥发油及P-糖蛋白(P-gp)抑制剂维拉帕米对3种人参皂苷类成分累积吸收量的影响。
结果:

缺失石菖蒲条件下定志小丸中3种人参皂苷类成分的

。与缺石菖蒲组相比,缺石菖蒲挥发油组的

无显著性差异,但定志小丸全方组的

提高了3.97~8.35倍,

提高了3.99~8.49倍;外翻肠囊研究表明定志小丸中石菖蒲挥发油能显著促进3种人参皂苷类成分在大鼠肠道的累积吸收量,但没有剂量依赖性。
结论:

石菖蒲挥发油可明显促进定志小丸中3种人参皂苷类成分的肠吸收,其促吸收机制可能与抑制P-gp有关。
Objective:

To explore the effect of Acori Tatarinowii Rhizoma on intestinal absorption of ginsenosides in Dingzhi Xiaowan
and reveal the mechanism of Acori Tatarinowii Rhizoma acting as " adjuvant drug" in this formula.
Method:

The contents of ginsenoside Rg
1
Re and Rb
1
were measured by UPLC-MS/MS and the absorption of three ginsenosides in different intestine segments was investigated by rat single pass intestinal perfusion
in situ
including absorption rate constant(
K
a
) and apparent permeability coefficient(
P
app
). Everted intestinal sac model was used to investigate the absorption dosage of three ginsenosides affected by volatile oil from Acori Tatarinowii Rhizoma and verapamil[Ver
a P-glycoprotein(P-gp) inhibitor].
Result:

values of three ginsenosides were ≤0.191×10

in Dingzhi Xiaowan when lack of Acori Tatarinowii Rhizoma.Compared with lack of Acori Tatarinowii Rhizoma in Dingzhi Xiaowan group

values of lack of volatile oil from Acori Tatarinowii Rhizoma in Dingzhi Xiaowan group slightly increased without significant difference in the four intestinal segments
but when the prescription had Acori Tatarinowii Rhizoma
the
K
a
increased by 3.97-8.35 fold and the
P
app
increased by 3.99-8.49 fold.The results of everted intestinal sac test showed that volatile oil of Acori Tatarinowii Rhizoma could significantly promote the intestinal absorption of ginsenoside Rg
1
Re and Rb
1
but there was no dose-dependent.
Conclusion:

Volatile oil of Acori Tatarinowii Rhizoma can promote the intestinal absorption of three ginsenosides in Dingzhi Xiaowan
and the mechanism may be related to the inhibiting function on P-gp.
