目的:

观察加味升降散对膜性肾病(MN)大鼠模型血液生化指标及对肾线粒体活性氧(ROS)表达的影响。
方法:

将60只SD雄性大鼠,适应性喂养7 d后随机分为4组,造模成功后西药组给予盐酸贝那普利(10 mg·kg
-1
·d
-1
)灌胃,中药治疗组给予加味升降散(13.22 g·kg
-1
·d
-1
)灌胃,模型组和正常组给予同等剂量的生理盐水灌胃。4周后,生化试剂盒测定各组大鼠24 h尿蛋白定量(UTP),血清总胆固醇(TC),甘油三酯(TG),总蛋白(TP),白蛋白(ALB),尿素氮(BUN),肌酐(SCr),实时荧光定量聚合酶链式反应(Real-time PCR)测定大鼠肾组织线粒体ROS mRNA的表达。
结果:

与正常组比较,其余各组造模后UTP,TC,TG均升高(
P

0.01),TP,ALB显著下降(
P

0.05);治疗后,加味升降散组、盐酸贝那普利组与正常组比较,血液生化指标及对肾线粒体ROS均改善,但未恢复正常水平。治疗结束后,与模型组比较,加味升降散组、盐酸贝那普利组UTP,TC,TG均有所降低(

0.05),加味升降散组、盐酸贝那普利组TP,ALB均有所升高(
P

0.05)。加味升降散组与盐酸贝那普利组UTP,TC,TG,TP,ALB比较无统计学意义。治疗结束后,与模型组比较,加味升降散组、盐酸贝那普利组大鼠肾组织中足细胞线粒体ROS mRNA的表达较模型组明显下调 (

0.05);加味升降散组与盐酸贝那普利组线粒体ROS mRNA比较差异无统计学意义。
结论:

加味升降散对膜性肾病大鼠的肾保护作用可能与其能够下调肾组织线粒体ROS mRNA表达,抑制足细胞凋亡、修复受损足细胞有关。
Objective:

To detect the effect of modified Shengjiangsan on the expression of reactive oxygen species(ROS) in mitochondria of renal foot cells of rats
in order to study the mechanism of modified Shengjiangsan.
Method:

After be fed for 7 days
the 60 SD male rats were randomly divided into four groups: blank control group
model group
positive medicine group and traditional Chinese medicine(TCM) treatment group. After establishment of the rat model of membranous nephropathy
model group
positive medicine group and TCM treatment group were treated differently. After 4 weeks
all of the rats were put to death
and the expressions of ROS
24-hour urinary protein quantity
total cholesterol
triglyceride
total protein
albumin
urea nitrogen
creatinine were detected by Real-time fluorescence quantitative polymerase chain reaction Real-time PCR.
Result:

The expression of 24-hour urinary protein quantity
total cholesterol
triglyceride in positive medicine group and TCM treatment group were reduced
and the expressions of total protein
albumin in positive medicine group and TCM treatment group were reduced compared with those of model group (
P

0.05). The expressions of 24-hour urinary protein quantity
total cholesterol
triglyceride
total protein
albumin in TCM treatment group have no difference with those of positive medicine group. After treatment
the expression of ROS mRVA in mitochondria of renal foot cells of rats of positive medicine group and TCM treatment group were increased compared with that of model group (

0.05)
but with no difference in the ROS mRNA expression between positive medicine group and TCM treatment group.
Conclusion:

Modified Shengjiangsan can effectively control the development of ROS in mitochondria of renal foot cells of rats
and repair the renal function of membranous nephropathy rats by recovering foot cells.
