目的:

观察酸枣仁汤对APP/PS1小鼠海马神经炎症的影响,并探讨其神经保护的可能机制。
方法:

将小鼠随机分为空白组,模型组,多奈哌齐组(0.92 mg·kg
-1
),酸枣仁汤低、高(12.96,25.92 g·kg
-1
)剂量组。各组连续给药30 d后,采用尼氏染色观察各组小鼠海马齿状回(DG)区病理形态的变化,采用酶联免疫吸附测定(ELISA)检测各组小鼠血清中肿瘤坏死因子-
α
(TNF-
α
)和白细胞介素-1
β
(IL-1
β
)表达水平,采用实时荧光定量聚合酶链式反应(Real-time PCR)检测各组小鼠海马中TNF-
α
,IL-1
β
的mRNA表达水平,采用免疫组化(IHC)和蛋白免疫印迹法(Western blot)检测各组小鼠海马中胶质纤维酸性蛋白(GFAP)和离子钙结合蛋白(IBA1)蛋白表达水平。
结果:

与空白组比较,模型组小鼠海马DG区颗粒细胞排列不均匀,细胞丢失明显,部分神经元胞内尼氏体消失或固缩,血清中TNF-
α
,IL-1
β
含量显著升高(

0.01),小鼠海马中TNF-

mRNA表达显著升高(

0.01),海马DG区中GFAP,IBA1蛋白表达显著上调(

0.01);与模型组比较,多奈哌齐组、酸枣仁汤低、高剂量组小鼠海马DG区颗粒细胞排列稍整齐,神经元丢失减轻,且胞内尼氏体消失或固缩情况好转,小鼠血清中TNF-

酸枣仁汤能改善APP/PS1双转基因小鼠神经元丢失,其机制可能与其调节小鼠海马神经炎症有关。
Objective:

To observe the effect of Suanzaoren Tang on hippocampal neuroinflammation in APP/PS1 mice and to explore its possible mechanism of neuroprotection.
Method:

The mice were randomly divided into blank group
model group
donepezil group(0.92 mg·kg
-1
)
Suanzaoren Tang low and high-dose groups(12.96
25.92 g·kg
-1
). After 30 days of continuous administration in each group
pathological changes of dentate gyrus (DG) in hippocampus of mice in each group were observed by Nissl staining.The levels of tumor necrosis factor-
α
(TNF-
α
) and interleukin-1
β
(IL-1
β
) in serum of each group were detected by enzyme-linked immunosorbent assay (ELISA). Real-time quantitative polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression levels of TNF-
α
and IL-1
β
in hippocampus of each group. The expression levels of glial fibrillary acidic protein (GFAP) and ionic calcium binding protein 1 (IBA1) in hippocampus of each group were detected by immunohistochemical staining (ICH) and Western blot.
Result:

Compared with blank group
the granule cells in DG region were unevenly arranged in model group
with obvious cell loss
and the nissl bodies in some neurons disappeared or condensed
serum TNF-

and IL-1
β
content significantly increased (

Suanzaoren Tang can improve neuronal loss in APP/PS1 double transgenic mice
and its mechanism may be related to the regulation of hippocampal neuroinflammation in mice.
