建立当归不同品种间差异代谢物,为当归新品种育种、引种、区划栽培和生态种植提供参考依据。
方法:

对同一产地、同期采收的5个当归品种岷归1号(MG1),岷归2号(MG2),岷归4号(MG4),岷归5号(MG5)和岷归6号(MG6)进行非靶向代谢组学综合分析。取各品种药材50%甲醇提取物,采用超高效液相色谱-四极杆-飞行时间质谱(UPLC-Q-TOF-MS)技术,结合软件Progenesis QI,通过主成分分析(PCA)和偏最小二乘判别分析(PLS-DA)等方法找出各品种间差异代谢物;依据相对分子质量,二级碎片,KEGG数据库和HMDB数据库以及相关文献信息,对差异代谢物进行鉴定。
结果:

5个当归品种的代谢物存在明显差异(
P

0.05),MG2,MG4,MG5和MG6中的绿原酸、阿魏酸、色氨酸、阿魏醛等含量显著低于MG1含量(
P
<
0.05),而藁本内酯、香豆素、牛角苷、棕榈素、原儿茶醛、亚麻酸等含量显著高于MG1(
P
<
0.05)。MG2,MG5及MG6中的代谢物相近,与MG4的代谢物差异较大,此外,还鉴定出38个显著差异代谢物,涉及7种潜在的靶向代谢通路,不同品种当归都可能通过苯丙素类生物合成、类倍半萜烯类化合物的代谢、脂质代谢、氨基酸代谢、碳水化合物代谢、核苷酸代谢、类胡萝卜素、亚麻酸、亚油酸等代谢途径调节代谢物的合成。
结论:

应用UPLC-Q-TOF-MS和Progenesis QI代谢组学技术从整体水平比较不同品种当归的化学成分,发现其中的差异性及其规律,为当归质量控制、引种、栽培及当归育种和生态种植提供参考依据。

Method:

Comprehensive non-target metabonomics analysis was conducted for five new varieties of
A
.
sinensis
collected at the same time from the same origin: Mingui No. 1 (MG1)
Mingui No. 2 (MG2)
Mingui No. 4 (MG4)
Mingui No. 5 (MG5)
and Mingui No. 6 (MG6). The 50% methanol extract of each variety was taken
and then the differential metabolites among varieties were found by using ultra-high performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS)
software Progenesis QI
principal component analysis (PCA)
partial least squares discriminant analysis (PLS-DA)
and non-targeted metabonomics analysis. Differential metabolites were identified based on precise molecular weight
secondary fragments
KEGG database
HMDE database and related literature information.
Result:

The results of UPLC-Q-TOF-MS and multivariate statistical analysis showed that there were significant differences in the metabolites of five
Angelica
varieties. As compared with MG1
the contents of chlorogenic acid
ferulic acid
tryptophan and ferulic aldehyde were significantly lower in MG2
MG4
MG5 and MG6
while the contents of ligustilide
coumarin
bovine keratin
palmitin
protocatechualdehyde and linolenic acid were significantly higher (
P
<
0.05). The results of multivariate statistical analysis showed that the metabolites of MG2 and MG5 were similar with those of MG6
but were significantly different from those of MG4.In addition
38 distinct metabolites were identified
involving 7 potential targeted metabolic pathways. Different varieties of
A
.
sinensis
could regulate the synthesis of their metabolites through phenylpropanoid biosynthesis and sesquiterpene-like compounds metabolism
lipid metabolism
amino acid metabolism
carbohydrate metabolism
nucleotide metabolism
carotenoids
linolenic acid
linoleic acid and some other metabolic pathways.
Conclusion:

UPLC-Q-TOF-MS and Progenesis QI metabonomics techniques were used to compare the chemical constituents of different varieties of

sinensis
