目的:

比较3种冰片不同剂量防治给药改善急性心肌梗死(AMI)模型大鼠作用的强弱以及对氧化应激因子的影响,为阐明其抗心肌梗死量效关系及机制提供参考。
方法:

将健康成年SPF级SD雄性大鼠随机分为假手术组、模型组、溶剂模型组、硝酸甘油组、天然冰片高、中、低剂量(0.6,0.3,0.15 g·kg
-1
)组,艾片及合成冰片高、中、低剂量(0.2,0.1,0.05 g·kg
-1
)组,共13组,每组20只。按20 mL·kg
-1
灌胃,1次/d,连续预防给药3 d;假手术组与模型组给予同体积蒸馏水,溶剂模型组给予同体积5%聚山梨酯80。干预给药第3天,末次给药30 min后,施予冠状动脉左前降支结扎造模,将造模成功大鼠连续治疗给药3 d。采用BL-420N生物系统分析仪记录大鼠心电ST段波幅及血流动力学变化;大鼠体质量及心脏称重,计算心脏脏器系数;2,3,5-氯化三苯基四氮唑(TTC)染色计算心肌梗死率;苏木素-伊红(HE)染色评价心肌病理损伤程度;按试剂盒要求,检测血清中心肌酶乳酸脱氢酶(LDH),天门冬氨酸氨基转氨酶(AST),肌酸激酶同工酶(CK-MB)及氧化应激因子超氧化物歧化酶(SOD),丙二醛(MDA)的水平。
结果:

与假手术组比较,模型组大鼠ST段波幅于5 min后显著抬高,左心室舒张压(LVDP)值显著升高,左室心肌收缩成分实测最大缩短速度(Vpm)值显著降低,心脏脏器系数及心肌梗死率显著升高,心肌病理组织严重损伤,血清CK-MB,AST,LDH,MDA含量显著升高(

0.01)。与溶剂模型组比较,天然冰片及艾片中、低剂量,合成冰片高剂量均能显著抑制大鼠心电不同时间点ST段的异常抬高,天然冰片与艾片高、中剂量、低剂量及合成冰片高剂量均可显著升高左心室收缩压(LVSP)值,降低LVDP值(

3种冰片各剂量组能不同程度发挥心肌保护作用。此实验条件下改善心肌梗死药效呈艾片>天然冰片>合成冰片趋势,天然冰片量效呈负相关、合成冰片呈正相关,艾片无明显量效关系。
Objective:

The effects of three different doses of borneol on acute myocardial infarction (AMI) model rats and the effects on oxidative stress factors were compared to provide reference for elucidation of the dose-effect relationship and mechanism of anti-myocardial infarction.
Method:

Healthy adult male SPF SD rats were randomly divided into sham operation group
model group
solvation model group

a total of 13 groups
20 in each group. Gavage was performed at 20 mL·kg
-1
once a day for 3 days of continuous preventive administration. The sham operation group and the model group were given the same volume of distilled water
and the solvation model group was given the same volume of 5% polysorbate 80.On the third day of the pre-administration

the left anterior descending coronary artery was ligated to make a model
and the successful rats were treated for 3 days. BL-420N biological system analyzer was used to record the ST-segment amplitude and hemodynamic changes. Rat body weight and cardiac weight were weighed to calculate cardiac viscera coefficients

5-triphenyl tetrazolium chloride (TTC) staining was used to calculate the myocardial infarction rate. Hematoxylin-eosin (HE) staining was used to evaluate the degree of myocardial pathological damage. According to the kit requirements
serum levels of lactate dehydrogenase (LDH)
aspartate amino-transaminase (AST)
creatine kinase isoenzyme (CK-MB) and oxidative stress factors superoxide dismutase (SOD)
malondialdehyde (MDA) were detected.
Result:

Compared with the sham operation group
the ST segment amplitude of the model group significantly increased after 5 minutes
the left ventricular diastolic blood pressure (LVDP) value increased significantly
and the measured maximum shortening velocity (Vpm) value of the left ventricular myocardial contraction component significantly decreased. The organ coefficient and myocardial infarction rate were extremely significantly increased
and the myocardial pathological tissue was severely damaged. The serum CK-MB

Three kinds of borneol in different dose groups can play different degrees of myocardial protection. Under the experimental conditions
there was a trend of l-Borneolum>Borneolum>Borneolum syntheticum in improving the efficacy of myocardial infarction
the dose-effect of Borneolum was negatively correlated
Borneolum syntheticum was positively correlated
and no significant dose-effect relationship between l-Borneolum.
