目的: 研究利心冲剂对心衰大鼠神经内分泌因子及炎性相关因子的影响。 方法: 实验分组:正常对照组、模型组、利心冲剂低剂量组(20 g ·kg-1)、利心冲剂高剂量组(40 g ·kg-1)、卡托普利阳性药组(16 mg ·kg-1 )。ip阿霉素(ADR)4 mg ·kg-1

形态学显示心肌细胞变性坏死明显减轻;左心室收缩内压显著升高
以高剂量最为明显(P<0.01);ADR作用后心肌细胞神经内分泌因子相关指标BNP
ANP与ETmRNA明显升高(P<0.01)
利心冲剂治疗后BNP
ANP与ETmRNA明显降低
以高剂量最为明显(P<0.01);ADR作用后心肌细胞炎性相关因子TNF-α明显升高(P<0.01)
利心冲剂治疗后TNF-α明显降低
以利心冲剂高剂量组最为明显(P<0.01);ADR作用后心肌细胞TIMP-1表达明显减少(P<0.01)
利心冲剂治疗后TIMP-1表达明显增加
以高剂量最为显著(P<0.01)。 结论: 利心冲剂调控神经内分泌因子及炎性相关因子
改善心脏功能。
Objective: To observe the protective effect of Lixin granule on the neuroendocrine and inflammation of rats with adriamycin-induced heart failure. Method :Sixty adult male Wistar rats were divided into 5 groups:normal control group(NC)
adriamycin(ADR)group
L- Lixin granule group(20 g ·kg-1)
H- Lixin granule group(40 g ·kg-1)
captopril group(16 mg ·kg-1 ). Then ADR of 4 mg ·kg-1 was given intraperitoneally once in every 5 days(total 3 times)to copy the model of heart failure. The biological signal collecting system was used to record and analyze the LVSP of the rats. The pathological changes in the cardiomyocytes were observed. Real time PCR was used to record and analyze the neuroendocrine and inflammation of rats. Immunohistochemistry staining was used to detect TIMP-1. Result: As compared with NC group
the LVSP of the ADR group was significantly lower(P <0.05)
but those of the H- Lixin granule group were markedly higher than those of the ADR group(P <0.01). HE staining showed that the myocardial tissue in CHF group was injured
presenting in cardiomyopthy-like changes;in NC group
muscle fiber was arranged regularly with no injury muscle fibers;in the treatment group
Lixin granule partially normalized contractile function. Real time PCR and Serological detection staining showed that the neuroendocrine factors(ANP
BNP
ET)and inflammatory factor(TNF-α)of rats were increased by ADR
and those of the H- Lixin granule group were markedly lower compared with ADR group(P<0.01). Conclusion: H- Lixin granule group treatment can relieve the neuroendocrine and inflammation induced by ADR in myocardium
and also obviously improve the cardiac contractility of heart failure rats.
