Objective: To explore the anti-atherosclerosis (AS) effect of serum containing Tanyu Tongzhi Fang(TYTZF) on the activation of nuclear factor-kappa B(NF-κB) and the expression of intercellular adhesion molecule-1(ICAM-1) in human umbilical vein endothelial cells (HUVECs) injured by oxidized low density lipoprotein (ox-LDL). the serum was taken for testing. HUVECs were treated with serum containing TYTZF and simvastatin respectively and incubated with ox-LDL (100 mg·L-1) for an additional 3 h or 24 hours. The mRNA levels of NF-κB p65 and ICAM-1 were measured by real-time PCR and the protein expression of ICAM-1 was detected by Western blotting. The activation of NF-κB p65 was observed with immunofluorescence method. Result: The expression of NF-κB p65 and ICAM-1 in serum containing TYTZF group and simvastatin group were significantly lower than that in modle group (P<0.05)
especially in the high-dose serum containing TYTZF group and simvastatin group(P<0.01). Conclusion: The mechanism of TYTZF anti-AS may be related to the inhibitory effect of NF-κB pathway
thereby reducing the expression of ICAM-1 in vascular endothelial cells.
