摘要

目的: 优选知母皂苷AⅢ的酶解工艺。方法: 采用β-葡萄糖苷酶水解知母皂苷制备知母皂苷AⅢ

以知母皂苷AⅢ转化率为指标

通过单因素和正交试验考察反应温度、反应时间、pH及酶用量对知母皂苷AⅢ酶解工艺的影响。结果: 最佳酶解工艺条件为温度55℃

pH 5.0乙酸-乙酸钠缓冲液

酶用量600 U·g-1

反应时间 3 h。结论: 优选的酶解工艺简单可靠

反应条件温和

适用于工业化生产。

Abstract

Objective: To optimize enzymatic technology of timosaponin AⅢ. Method: Taking bioconversion rate of timosaponin AⅢ as index

β-glycosidase enzyme was used to hydrolysis timosaponin

effect of reaction temperature

reaction time

pH and dosage of enzyme on enzymatic technology of timosaponin AⅢ was investigated by single factor test and orthogonal test. Result: Optimum enzymatic technology conditions were as follows:reaction temperature of 55℃

pH of acetic acid-sodium acetate buffer solution 5.0

dosage of enzyme 600 U· g-1

reaction time 3 h. Conclusion: Optimized enzymatic technology was simple and reliable with moderate reaction conditions

it was suitable for industrial production.
