目的: 探讨经方茵陈蒿汤对高脂饮食诱导大鼠非酒精性脂肪性肝炎(non-alcoholic steatohepatitis
NASH)的药效作用。 方法: 以高脂饲料喂养SD雄性大鼠8 周建立NASH大鼠模型
随机分为模型组、茵陈蒿汤高剂量组、茵陈蒿汤低剂量组
复方益肝灵组及自愈组。茵陈蒿汤高、低剂量组、复方益肝灵组于第9周起分别给予茵陈蒿汤高剂量4.97 g·kg-1、低剂量3.31 g·kg-1、复方益肝灵1.265×10-2 g·kg-1治疗4周
自愈组恢复正常饮食。4周后观察药物对大鼠血清脂质、肝脏脂质、血清胰岛素含量及抗氧化指标的影响
对肝组织进行常规HE、油红O染色。 结果: 茵陈蒿汤低剂量能明显降低模型大鼠血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)
血清和肝内丙二醛(MDA)、肝脏游离脂肪酸(FFA)含量
增加血清谷胱甘肽过氧化物酶(GSH-Px)活性、降低血清胰岛素(Ins)含量(P<0.05
P<0.01)
改善高胰岛素血症和肝组织脂质沉积的病理状态。茵陈蒿汤高剂量对血清TG
MDA
GSH-Px
Ins
肝内FFA部分指标有所改善(P<0.05
P<0.01)。 结论: 茵陈蒿汤低剂量对改善大鼠NASH的作用优于高剂量。茵陈蒿汤低剂量能够有效针对非酒精性脂肪性肝病(non-alcoholic fatty liver disease
NAFLD)发病的"二次打击"机制调节高脂饮食诱导的大鼠脂质代谢紊乱和高胰岛素血症状态
增强抗氧化能力、清除自由基代谢产物
抑制脂质过氧化反应
从而达到治疗NASH的作用。
Objective: To observe the effects of Yinchenhao Decoction(YCHD) on non-alcoholic steatohepatitis (NASH) rats induced by high fat diet. Method: NASH model in SD rats was established by feeding high fat diet for 8 weeks
and then the rats were randomized into model group
YCHD high dose group
YCHD low dose group
composite prescription Yiganling group
natural recovery group. Rats in medication groups were administrated ig with corresponding herbs from the 9th week
YCHD groups (4.97

the influence of the treatments on blood lipids
liver lipids
antioxidant indexes and serum insulin contents were investigated. Hepatic tissues were detected by HE and oil red staining. Result: The level of total cholesterol(TC)
triglycerides(TG)
low density lipoprotein(LDL-C)
malondialdehyde(MDA)
insulin(INS)
free fatty acids(FFA)
MDA in liver of YCHD low dose group were significantly lower than those of model group. The activities of glutathine peroxidase (GSH-Px) was significantly higher than that of model group(P<0.05
P<0.01). It can improve the steatosis of liver in rats with NASH.Meanwhile
the level of TG
Ins
MDA in serum and FFA in liver of YCHD high dose group were also significantly lower than those of model group(P<0.05
P<0.01). Conclusion: YCHD low dose is better than high dose on rats NASH model. YCHD low dose has good effects on NASH rats feeding by high fat diet with disorders of lipid metabolism and hyperinsulinemia for the pathogenesis of NAFLD ‘two-hit’ mechanism by enhancing antioxidant ability and clearing free radical metabolites as well as inhibiting lipid peroxidation.
