摘要

目的：研究温中益气胶囊对幽门结扎型胃溃疡大鼠的治疗作用以及对小鼠的抗炎、镇痛作用。方法：采用幽门结扎法复制大鼠胃溃疡模型。取清洁级健康SD大鼠60只，雌雄各半，随机分成空白组，模型组，阳性对照药三九胃泰颗粒组（3.69 g·kg-1），温中益气胶囊高、中、低剂量组（2.21，1.11，0.55 g·kg-1）。每天ig给药1次，连续给药7 d，模型组和空白组给予等量蒸馏水。第5天给药后各组动物禁食不禁水48 h，末次给药后2 h麻醉，进行幽门结扎手术，缝合后直肠给药1次，禁食水。18 h后麻醉大鼠，颈总动脉取血。取全胃观察大鼠胃溃疡面积和溃疡指数，胃组织匀浆测定丙二醛（MDA）、超氧化物歧化酶（SOD）、H+-K+-ATP酶、生长抑素（SS）含量。采用二甲苯致小鼠耳廓肿胀和热板刺激法进行抗炎和镇痛实验，测定小鼠耳肿胀度、抑制率及痛阈值的大小。分别取清洁级小鼠50只，随机分成空白组、阳性对照药三九胃泰颗粒组（5.19 g·kg-1）、温中益气胶囊高、中、低剂量组（3.12，1.56，0.78 g·kg-1）。结果：胃溃疡实验中，与空白组比较，模型组H+-K+-ATP酶活性和MDA含量显著增加（P<0.01），SS和SOD活性显著降低（P<0.05，P<0.01）；与模型组比较，温中益气胶囊能显著减小大鼠胃溃疡面积和溃疡指数，降低H+-K+-ATP酶的活性和MDA含量并增加胃组织中SS和SOD水平（P<0.01，P<0.05）。抗炎、镇痛实验中，与空白组比较，温中益气胶囊能降低二甲苯致小鼠耳廓肿胀度，提高小鼠痛阈值（P<0.01或P<0.05）。结论：温中益气胶囊可能通过降低H+-K+-ATP酶活性和MDA含量、增强SOD活性、增加SS的含量来实现抗幽门结扎型胃溃疡作用，并具有明显的抗炎、镇痛作用。
