目的:确认白芍改善睡眠作用的药效物质基础,为揭示其改善睡眠作用的机制提供可靠依据。 方法:Wistar大鼠随机分为正常对照、芍药苷12 mg·kg-1,给药组ig(灌胃)给药,每天1次,连续7 d,于末次给药后5 h制备血清或抽取脑脊液,用HPLC测定血清和脑脊液中四逆散移行成分;或者连续ig给药7 d后,ip给予戊巴比妥钠50 mg·kg-1,用翻正反射法测定小鼠睡眠时间。采用HPLC方法,梯度洗脱,色谱条件为:Hypersil C18柱(4.6 mm×250 mm,5 μm),流速0.8 mL·min-1,柱温25℃,流动相A为乙腈;流动相B为(pH 3)磷酸水,流动相A梯度洗脱(5%~95%乙腈),分析时间为140 min。 结果:12 mg·kg-1芍药苷可以使脑脊液中内源性物质峰面积高于空白脑脊液峰面积,从峰面积比值来看芍药苷能使脑脊液中内源性物质面积约是空白脑脊液峰面积的10.2倍。表明白芍主要化学成分芍药苷能明显促进脑脊液中内源性物质的分泌,从而达到改善睡眠作用。 结论:通过白芍血中移行成分分析、脑脊液中移行成分分析及白芍对戊巴比妥钠所致小鼠睡眠时间影响的药效研究,确认了白芍改善睡眠作用的药效物质基础是血清中白芍的移行成分芍药苷。
Objective: To study active constituents of Paeoniae Radix Alba with improving sleep function. Method: Wistar rats were randomly divided into normal control
paeoniflorin 12 mg·kg-1
administered group(intragastric administration) ig administered once daily for 7 d
after the last administration of the preparation of serum or cerebrospinal fluid extraction
and serum by HPLC Sini San transitional cerebrospinal fluid composition;after ig administration or continuous 7 d
ip sodium pentobarbital 50 mg·kg-1
sleep time was measured using mouse righting reflex method.By using HPLC method
the gradient elution
and determine the transmigration ingredient of blood serum in Paeoniae Radix Alba. The chromatograph condition:Hypersil C18 column (4.6 mm×250 mm
5 μm)
speed of flow 0.8 mL·min-1
column warm 25℃
flowing A is acetonitrile;flowing B is(pH 3)phosphorus acid water
the flowing A gradient elution (5%-95% acetonitrile)
the analysis time is 140 min. Result: By the analysis of the compound in cerebrospinal fluid
we found that there was no other compound absorbed into blood except for the pentobarbitalum natricum. Moreover
the endogenous substance in cerebrospinal fluid was increased obviously. Increasing peak area of the endogenous substance in cerebrospinal fluid. Results of the pharmacological experiment indicated that Paeoniae Radix Alba significantly prolonged the sleep time of mice induced by pentobarbital sodium. Conclusion: Finally
definiting active constituents of Paeoniae Radix Alba improving sedation function
confirmed the efficacy of the material basis of the Paconiae Radix Alba is paeoniflorin in the serum.
